SDRT:
The Science

  1. A temporary disruption of tumor blood flow results in an acute reduction of oxygen perfusion throughout the tumor tissue (“ischemic phase hypoxia”)
     

  2. Hypoxia injures the mitochondrial respiratory chain system in tumor cells and inevitably causes diffuse damage to the metabolic function of the cell via generation of reactive oxygen species (ROS)
     

  3. In response, the tumor cells activate a rescue operation, termed “SUMO Stress Response” (SSR)
     

  4.  An element of SSR is transient silencing of normal cellular functions, such as the assembly and activation of homology-driven repair (HDR) of DSBs, an adaptive cell lethality function activated by exposure to SDRT
     

  5. A global loss of both HDR and repair of DSBs leads to extensive DNA disintegration in tumor cells, generalized tumor cell death, and tumor ablation
